Natreon’s PrimaVie Collagen Gene Expression Study published

A NEW HUMAN CLINICAL STUDY from The Ohio State University Wexner Medical Center, published in PubMed listed Journal of Medicinal Food, shows that oral supplementation with Natreon’s PrimaVie® Purified Shilajit can significantly up-regulate the genes responsible for synthesis of collagen and other extracellular matrix proteins resulting in adaptation of the skeletal muscle mechanotrasduction properties, elasticity, repair and regeneration.

Natreon’s PrimaVie®, a high quality, purified and standardized extract of Shilajit, significantly up-regulated the genes responsible for collagen synthesis according to a human clinical study just published in the Journal of Medicinal Food. PrimaVie®, a GRAS affirmed and patented ingredient, is comprised of mainly fulvic acids and DBPs (dibenzo-α-pyrones). Fulvic acid independently stimulates mitochondrial energy metabolism, protects mitochondrial membranes from oxidative damage, and helps channel electron-rich DBPs into the mitochondria to support the electron transfer chain. Several clinical studies have shown that PrimaVie® can boost mitochondrial energy, improve exercise performance, and increase testosterone levels. In this open label longitudinal study, sixteen overweight/Class I obese Americans (both genders) were followed for 12 weeks to determine the effects of oral supplementation of PrimaVie® and exercise training on human skeletal muscle adaptation. This particular cohort was selected for the study because physiological muscle performance is often compromised in this group of subjects. The subjects received 250 mg of PrimaVie® twice daily for 8 weeks followed by an additional 4 weeks of supplementation with exercise. To determine the changes in mRNA expression profile in response to PrimaVie® supplementation and exercise, skeletal muscle biopsies of the thigh muscle were done followed by RNA extraction, target labeling, and GeneChip data analysis. The genes that were expressed highly in skeletal muscle samples after 8 weeks of supplementation compared to baseline included tenaxcin XB (TNXB), decorin (DCN), myoferlin (MYOF), collagen (COL) (type I, II, III, V, VI, XIV), elastin (ELN), fibrillin 1(FBN1), and fibronectin 1 (FN1). Several collagen related genes, including COL1A1, COL1A2, and COL3A1 were increased by 4.61 (p = .0149), 5.13 (p = .0076), and 5.18 fold (p = .0086), respectively. To validate the genes identified using microarray analysis, RT-PCR was performed and the up-regulated genes mentioned above were re-examined. Consistent with microarray results, significant up-regulation of collagen and other ECM-associated genes was noted in the muscle samples (some up to 1500 fold) in the muscle samples following 8 weeks of oral supplementation of PrimaVie® compared with the baseline visit. Interestingly, an additional 4 weeks of PrimaVie® supplementation with exercise further induced the expression of the microarray identified genes. In addition, PrimaVie® supplementation was well tolerated without any changes in blood glucose levels and lipid profile. Although additional studies will follow to confirm the effects of oral supplementation of PrimaVie®, the increase in collagen expression in this study suggests the potential role of PrimaVie® Purified Shilajit in healthy aging.

New highlights on folate MTHFR polymorphism from ICHOCM 2017. Really interesting scientific data have been presented at the just closed 11th  International Conference on Homocysteine and One-Carbon Metabolism, held in Aarhus, Denmark last week. During the four-day convention, many internationally experts gave lectures about the One-Carbon Metabolism (often referred as methylation cycle) and its link with Folate, SAMe and Glutathione, in order to share and learn the latest scientific knowledge and experiences and enhance our facilities.

Among others, two special focuses have been devoted to:

1) the role of methyltetrahydrofolate reductase polymorphism (MTHFR) and its incidence in health and disease, including cardiovascular disease, neurodegenerative disease (Alzheimer’s disease and dementia, Parkinson’s disease), birth defects, and comorbidities associated with obesity;

2) challenges to evaluating evidence of risks from high intakes of folic acid by supplementation.

MTHFR enzyme catalyzes the irreversible conversion of 5,10-methylenetetrahydrofolate to the active form of folate, the 5-methyltetrahydrofolate (5-MTHF) and is considered the key in the One-Carbon Metabolism pathway. Deficiencies in production or function due to polymorphism are implicated with elevated homocysteine concentrations and consequently in chronic disease states.

MTHFR deficiency increases the risk of stroke, reports the work presented by Dr. Nafisa M. Jadavji et al. Stroke is a leading cause of disability and death world-wide. She has investigated the mechanism through which the MTHFR polymorphism may have a role using in vitro and in vivo tools. Study results point out that individuals with the homozygous polymorphism of MTHFR (TT genotype) have elevated homocysteine concentrations and that MTHFR deficiency increases vulnerability to stroke and leads to more severe impairment after stroke through increased apoptosis and oxidative stress.

Regarding folic acid scientific research is increasingly interested on the potential for adverse effects of high intakes of folic acid. Interesting results have been presented by Dr. Emily C. McGowan et al. on the hypothesis that the increased food allergy prevalence observed in recent years may involve changes in our nutrition, including increased exposure to folic acid through fortification and supplement use.

Moreover new highlights have been presented about the connection between One-Carbon’s perturbations and the deficiency of specific nutrients vital for life such as S-adenosylmethionine/SAMe, and glutathione, both present in Gnosis’ pipeline like Adonat® and Emothion™.

There was no such increase in the placebo group. Abdominal muscle thickness increased in both groups, but the increase was 1.8 times greater in the Glavonoid group than in the placebo group (p<0.05). It can therefore be seen that Glavonoid™ is ideal for inclusion in products aimed at the sports and fitness market.

Combination of Natural-derived Vitamin K2 and Policosanol is effective in prevention of Cardiovascular Disease

The combination of natural-derived vitamin K2 with octacosanol (the main constituent of policosanol) could contribute to lowering levels of cholesterol, triglycerides, LDL-cholesterol, and its combination with a statin can achieve the target value of LDL- and HDL-cholesterol, say a new study published by Jovanovic et al in International Journal of Pharmaceutical Science and Invention.

The study was conducted as a prospective clinical study and included a total of 6595 subjects. The study group included 4031 subjects (1785 males and 2246 females) who were taking a combination of natural Vitamin K2/MK7 from Bacillus subtilis Natto 45μg , vitaMK7® (by Gnosis, Italy) with 20 mg of Octacosanol under the brand name of ARTEROprotect® (by Abela Pharm, Serbia) while the control group compromised 2564 subjects (1135 males and 1428 females) who were not taking it.

The study was conducted by 76 doctors – general practitioners, specialists in internal medicine and cardiology – in in primary health centers throughout the Republic of Serbia, in accordance with the principles of good clinical practice and the Declaration of Helsinki.

Based on the results, it can be said that dietary supplement ARTEROprotect® used alone can contribute to lowering levels of blood fat, and in combination with a statin it can achieve the target value of HDL-cholesterol.

The Wingate test was used to assess maximum anaerobic power during a high intensity

anaerobic work capacity by 5% which represents an increase of 876 joules developed in
30 seconds. 900 mg of PerfLoad®, taken pre-workout, was proven to provide explosive
power from the very start of the exercise and the maximal peak power output was
significantly increased by 3.7% without inducing higher fatigue or higher heart rate at
V02max. Besides, blood markers showed that oxidative homeostasis was stabilized in
supplemented subjects.
 
As pre-workout supplementation has become a significant nutritional approach in recent
years, such results demonstrated that the water-soluble ingredient, PerfLoad®, can be a
natural and efficient solution helping athletes to improve their physical performance
while balancing their metabolism and reducing exercise-induced oxidative stress.
